Efficiency of producing iPS cells markedly improved

first_imgSome of the most challenging obstacles limiting the reprogramming of mature human cells into stem cells may not seem quite as daunting in the near future. Two independent research groups, one lead by Harvard Stem Cell Institute Principal Faculty member Konrad Hochedlinger, and another at the Whitehead Institute, are describing new tools that provide invaluable platforms for elucidating the molecular, genetic and biochemical mechanisms associated with reprogramming. The work of both groups is being published in the September 11th issue of the journal Cell Stem Cell.Although scientists have successfully reprogrammed mature human skin cells into induced pluripotent stem cells (iPS) by expressing a few key transcription factors, the conversion has been extremely inefficient. “Little is known about the mechanisms by which reprogramming occurs, in part because of the low efficiency,” says Hochedlinger. In addition, the iPS cells created thus far have been generated with retroviruses and non-inducible lentiviruses, both of which have limitations that may limit clinical applications.The Hochedlinger group created a drug-inducible lentiviral system to generate human iPS cells that were molecularly and functionally similar to human embryonic stem cells. The researchers found that once the “primary” iPS cells differentiated to mature cells, addition of the drug doxycycline reactivated the genes required for reprogramming and induced generation of “secondary” iPS cells at a frequency that was far greater that the initial “primary” conversion. “The secondary system will enable chemical and genetic screening efforts to identify key molecular constituents of reprogramming, as well as important obstacles in this process, and will ultimately lend itself as a powerful tool in the development and optimization methods to produce human iPS cells,” explains Hochedlinger, an assistant professor in Harvard’s Department of Stem Cell and Regenerative Biology.A separate team of scientists, led by Rudolf Jaenisch of the Whitehead Institute and MIT, had parallel success in deriving human secondary iPS cells using doxycycline-inducible transgenes.  “The drug-inducible system we describe represents a novel, predictable, and highly reproducible platform to study the kinetics of iPS cell generation,” says Dr. Jaenisch.Both research teams found that generation of secondary human iPS cells required less time than the initial reprogramming. Interestingly, the time required to generate iPS cells varied among the types of skin cells that were used. For instance, human fibroblasts required several weeks while keratinocytes required only about ten days. “The fast kinetics of reprogramming observed for keratinocytes suggests that these cells would be useful for development and optimization of methods to reprogram cells by transient delivery of factors,” suggests Hochedlinger.The combined results from both research groups represent a major advance towards more efficient strategies for reprogramming differentiated human cells into iPS cells. Mertens pledges future to Inter?

first_imgThere are reports Napoli striker Dries Mertens has already agreed to join Inter as a free agent next summer for a salary of €6m per season. According to Sunday’s edition of the Corriere dello Sport and Gazzetta dello Sport, the Belgian has worked out basic personal terms to remain in Serie A, albeit at San Siro with a Nerazzurri jersey. It’d be a two-year contract with option for a third, worth €6m per season, including various bonuses. As he’ll be 33 by the end of this season, the deal could also include provisions for a future spell at Inter’s sister club Jiangsu Suning in China. Mertens has been at Napoli since 2013 when he arrived from PSV Eindhoven, but his contract is due to expire in June 2020. President Aurelio De Laurentiis has repeatedly suggested he does not want to keep Mertens on his current salary and would only sign him – and Jose Callejon – to a new deal at a vastly reduced rate. Mertens will be free to start negotiating a contract with another club from January and the Corriere dello Sport maintains he has already been approached by Antonio Conte’s Inter. With that in mind, Napoli could try to sell Mertens in the January transfer window to at least make back some money. The Tiger effect Woods presence boosts business excitement

Tiger Woods’ impact is expected to extend past the score card when the world’s No. 1-ranked golfer plays in the Memorial Tournament this weekend in Dublin, Ohio.When Woods announced that he would be playing in Columbus, local-area businesses and the Muirfield Village Golf club, the host of the event, raised expectations for attendance and sales.“With Tiger coming back to the tour, I see an increase in business,” said Jon Leach, part owner of the Bogey Inn bar in Dublin.The Bogey Inn normally anticipates between 6,000 and 9,000 people for Memorial Tournament parties, and Woods’ participation will increase that number by 30 percent, Leach said.Some officials at the tournament are less certain about the effects.“We don’t know what impact (Tiger playing) is going to have yet simply because it is the first year that he has been here since his off-course life became more public,” said Tom Sprouse, director of communications for the Memorial.Other employees appear more confident.“When Tiger is in the field, there are definitely more people here. There’s going to be a lot more interest,” said Dick Murray, Memorial security guard. “I would assume that, that same level of excitement will be here.”Increased attendance cannot be confirmed because the tournament does not keep track of crowd numbers, Sprouse said.Murray said he doesn’t need to see the numbers to notice a difference and is sure “that once it was announced that Tiger was going to be here, ticket sales went up.”Woods’ presence not only affects the numbers, but the attitudes of the patrons.The younger kids are always a lot more excited when Tiger is in the field, said John McKitrick, who has been working with the tournament for four years.Despite the larger galleries that will follow Woods, the tournament does not make any security arrangements for the popular golfer, Sprouse said.The Bogey Inn, however, does make adjustments, increasing its staff when Woods plays, Leach said.With each day Woods tees off this weekend, large crowds both on the course and in local bars are likely to follow. read more
